Who Wins

Pre-6, Pyke + Soraka win the 2v2: your healing sustains Pyke through any poke Vayne and Twitch deal, and your E silence interrupts Vayne's E tumble-into-condemn combo and Twitch's E activation window.
Post-6, your R global heal becomes critical for keeping Pyke alive during his roams, but Vayne's W true damage and Twitch's E burst can chunk you down faster than you can heal yourself if you are caught without your Q healing.

Your best trading pattern is to use your Q on minions to restore your own health, then use your W on Pyke immediately after he takes damage from a trade — never use your W preemptively before damage lands, as the heal is wasted and you lose health from the W cost unnecessarily.

You should generally not roam in this matchup: Vayne and Twitch are both capable of punishing an unprotected Pyke, and your presence in lane is what makes the 2v2 favorable; however, if Pyke has a clear kill setup in mid or jungle and your R is available, using your R globally is always correct without leaving lane.
Your roam timing should be limited to using R reactively, not physically walking to another lane.

Never stand in a position where both Vayne and Twitch can reach you simultaneously — you are the primary kill target in this matchup because eliminating you removes all sustain from the lane, and your low mobility means a Twitch E slow into Vayne E condemn will kill you before Pyke can peel.
