Who Wins

Pre-6, Pyke + Soraka win the 2v2: your hook range forces Vayne and Twitch to play passively, and Soraka's healing sustains you through any poke they manage to land; at level 2 your Q + E combo is a genuine kill threat on either of them.
Post-6, your R execute becomes a serious problem for Vayne and Twitch who are both squishy, but Vayne's R invisibility and Twitch's Q make landing your hook harder, so you must create picks rather than straight 2v2 fights.

Your best trading pattern is to use your E dash to close distance and immediately follow with your Q hook to lock one target, then let Soraka's healing sustain you while you deal damage — never throw your Q from max range telegraphed, as both Vayne and Twitch have mobility to dodge it.

You should roam aggressively once you hit level 6: your R execute is one of the strongest roaming tools in the game, and Vayne + Twitch are both weak laners who cannot punish a roam effectively if the wave is pushed in.
Optimal roam timing is after a successful hook-kill or summoner burn in lane, immediately rotating to mid where your R execute can snowball the game.

Never use your E aggressively without your Q available — your E is your escape tool in this matchup, and if you dash in without your Q ready, Vayne's E can knock you away and Twitch's E slow will leave you stranded in their range with no way out.
