Who Wins

Pre-6, you win if you land your hook — Nami's bubble into your hook is a double CC chain that Yunara cannot survive, and Karthus has no escape to bail them out.
Post-6, the matchup becomes harder because Yunara's R gives her a dash that lets her reposition over walls, making your hook easier to dodge, but Karthus remains immobile and is always a valid hook target.

Hook Karthus, not Yunara — Yunara has more mobility tools and can kite, but Karthus is stationary and dies instantly to your hook into Nami's follow-up.
After the hook lands, stack your passive with autos and let Nami's empowered auto chain the CC so Yunara cannot safely disengage.

Roaming is risky here because Karthus's R is a global threat that punishes you the moment you leave lane — if you roam and Karthus ults your mid laner, you've traded nothing.
Only roam if the wave is pushed in and Karthus is low enough that he cannot safely R, otherwise stay and apply pressure in lane.

Do not blow your hook on Yunara when she has R available — she will dash away and you've wasted your primary engage tool while Karthus continues to poke freely.
Save your hook for Karthus or for moments when Yunara is overextended without R cooldown.
