Who Wins

Pre-6, you win the 2v2 with a level 2 all-in using E+Q before Rell can mount-engage, because your CC chain is faster and Samira immediately capitalizes on the stun.
Post-6, you continue to win because your R into Samira's R is a faster and more damaging sequence than Rell's R setup, provided you initiate before Rell can land her W.

Your best pattern is to E onto Twitch specifically, since he is the squishier and more immobile target, chain Q to extend the stun, and let Samira stack her passive and R.
Targeting Rell directly is riskier because she can absorb your CC with her own resistances and counter-engage.

You should roam aggressively after pushing the wave with Samira, particularly at level 6 when your R gives you a reliable engage tool in other lanes.
Twitch and Rell have no meaningful way to punish your roam quickly, and Samira can hold the lane short-term due to her wave-clear pattern.

Do not waste your E onto Rell when she has her W available, because she will dismount and absorb your engage while Twitch repositions with Q stealth.
Save your E for Twitch or for moments when Rell's W is on cooldown to ensure your CC chain actually results in a kill.
