Who Wins

Pre-6, this lane is losing for you: Thresh's hook range outpaces your Q poke, and if he lands a hook, Samira can immediately stack her passive and burst you before Alistar can respond.
Post-6, your R's spell shield on activation and Alistar's R make you significantly harder to kill, but Samira's R can still delete you if you're caught without your E up.

Your best pattern is to use E to block Thresh's Q, then immediately follow up with Q+W poke on Samira while Alistar looks for a W+Q engage — the spell shield removal of Thresh's hook is what enables the trade, not your damage alone.
Without E available, play passively and do not commit to extended trades.

Use your W to push the wave fast and create a permanent freeze-or-push dilemma for the enemy; Samira cannot match your wave clear and Thresh will be forced to stay to protect her or lose CS.
A shoved wave also reduces the window where Thresh's hook can catch you in open space.

Do not stand in a position where Thresh can hook you through minions while your E is on cooldown — this is the single most punishable mistake in this matchup and Samira will immediately R if you're hooked at low health.
Track your E cooldown and position behind the wave when it's down.
