Who Wins

Pre-6, Zilean + Samira have the edge — Zilean's Q poke is reliable and his E speed makes Samira's trades dangerous, while your W polymorph is strong but requires close range to land.
Post-6, your R becomes a critical counter to Samira's R, but Zilean's R directly cancels your kill attempts, so neither side has a clean advantage in all-ins.

Your best pattern is to use your E on Twitch to boost his auto range and damage, then immediately follow with your W polymorph on Samira when she dashes in — this disrupts her passive stacking and her R channel.
Save your R defensively for when Samira or Zilean commits to a kill on Twitch, as the knockup buys time for Zilean's R cooldown to matter.

You should not roam early — Twitch is immobile and dies quickly if left alone against Samira + Zilean.
If you do roam, only do so after a shove at level 6 when Twitch has his Q available to escape, and only if Zilean is visibly committed to the bot lane so he cannot follow.

Do not use your W aggressively on Samira unless you are confident it will land — missing it removes your primary peel tool and gives Samira a free window to dash onto Twitch.
Also avoid standing in Zilean's double bomb range while trying to set up trades, as getting stunned before you can polymorph Samira is a common way this lane falls apart.
