Scuttle Contest

At level 3 you win the scuttle fight decisively because your E outranges everything Kayn has at that stage, and landing it forces him to either burn Flash or take a full combo while you sustain with passive blobs.
At level 6, you win the 1v1 by opening with E to knock him up, immediately following with W and R to keep him airborne and unable to use Q dashes to escape; the key condition is landing E as the fight initiator, not as a follow-up, since Kayn's Q mobility makes him slippery if he gets the first move.

Kayn will almost always start on the side of the map where he can farm toward a specific form — expect him to path toward melee-heavy camps for Rhaast or toward ranged/mage camps for Shadow Assassin.
He will look to avoid direct confrontation early and instead farm to level 6 quickly, so anticipate him taking a full clear before looking for his first gank around the 4-5 minute mark.

You should invade Kayn at his second buff after he starts his clear, around the 3:15-3:45 window, because he will be low on health from clearing and has no ultimate to escape with yet.
Target the buff opposite his starting side — if he starts Red, invade Blue — and use your E through the wall to cut off his escape route, as he cannot dash through walls with Q at that point in the game.

Do not let Kayn freely farm into level 11 without contesting him, because a fully transformed Kayn — especially Rhaast — becomes significantly harder to kill due to his W healing scaling with your maximum HP, which directly counters your tankiness.
Avoid wasting your E in a gank right before you need to contest him at scuttle or dragon, as missing that ability removes your primary engage tool and he will simply out-sustain or disengage any fight you attempt without it.
