Pre-6, you have a real kill threat on Ryze if you can close the gap with W and force a melee trade, because his root requires him to land E first and your W dash can interrupt his setup if you time it well — at level 2 you spike hard with W available and can all-in before he has his full E-W-Q combo online.
However, Ryze's E-W root at level 3 is a genuine counter to your all-in, so if he lands it cleanly you will take heavy damage while immobilized and cannot sustain it all back immediately.

Post-6, Ryze's R does not make him unkillable in lane — it has a long channel and delay, so if you're already on top of him when he tries to use it you can kill him before the portal activates.
Your sustained DPS and passive healing give you a real edge in extended fights post-6, but you must respect his root chain because a full E-W-Q-E-Q combo while rooted will chunk you significantly even with your healing.

Your best trading pattern is to wait for Ryze to push the wave and overextend, then W dash onto him and force a sustained melee trade where your passive healing and attack speed outscale his burst — if he tries to E-W you, dodge the W with your W dash or sidestep before he can apply Flux, because without the root his damage output drops dramatically.
Avoid short trades where he pokes with Q and backs off; you need to commit fully and stay on him long enough for your healing to matter.

Do not let Ryze freely shove the wave and roam with R — apply constant pressure to force him to stay in lane, and if he channels R, immediately interrupt it by walking toward him or using W, since the channel is interruptible and losing his R means he loses his primary macro tool.
Freezing the wave near your side of the lane denies him safe E-Q waveclear and forces him into melee range to last-hit, which is exactly where you want him.
