Pre-6, you lose the level 3 window if Locke lands a full three-stack Q before you can activate W, because his E-W burst with stacks deals enough damage to chunk you before your passive healing matters — play defensively at level 3 and use W proactively to deny his Q stacking rather than reactively after he has already committed.
Post-6, his R slow and execution threshold are dangerous in all-ins, but if you force him to use R without full nail stacks pre-applied and you have W available, you can survive the threshold and win the extended fight through your passive sustain and Q damage.

Your best trading pattern is to bait his E blink with aggressive positioning, then immediately activate W when he arrives so he is forced to fight inside your mist or disengage — inside W you are untargetable from his Q follow-up and can freely land your full Q snip sequence and E-empowered attacks on him.
If he activates his W to absorb damage and heal, respect that he can recast it at will and do not dump your entire Q into him during his W window; instead, pressure him with E-empowered attacks and wait for his W to expire before committing your highest-damage snips.

You want a slow push building in your favor so the wave crashes toward Locke's tower, forcing him to stand in a predictable position where you can walk up and threaten W-Q trades without risking a gank while he has priority.
Avoid hard shoving the wave early, as Locke can use the resulting reset to roam or set up a dive with his E reset mechanic, and you have limited tools to match his roam speed or follow effectively.

Do not use W reactively after Locke has already landed his E on you — the mist only blocks external targeting, and he is already inside melee range dealing damage, so you gain far less value than if you had placed W preemptively to deny his Q stacking approach.
Burning W as a panic response means you have no protection against his R slow and execution threshold in the same trade window, which is when you are most vulnerable.
